PRRS NA/HP detection kit Introduction

I.               Intended use

The ADNUCLEIS North America Porcine Reproductive and Respiratory Syndrome (PRRS) virus (HQS_PRRS-NA/HP_i_tqm) kit uses real time reverse transcriptase polymerase reaction (rt RT-PCR) amplification and is designed for the qualitative detection of North America Porcine Reproductive and Respiratory Syndrome virus from a RNA nucleic acid extract. This kit also contains an internal positive control (IPC) of RT-PCR. The kit allows the early diagnosis of presumption of the North America PRRS virus from semen.

II.               Introduction

Porcine reproductive and respiratory syndrome (PRRS) is the most costly viral disease in the U.S. and around the world (Holtkamp et al.,2013). PRRS affects both the reproductive and grow-finish sectors of the pork industry through a decrease in reproductive health, an increase in mortality and morbidity, and reductions in the rate and efficiency of growth (Zimmerman et al., 2012). PRRS virus (PRRSV) also functions as a cofactor in other disease syndromes, such as porcine respiratory disease complex (PRDC) and PCV-associated disease (PCVAD) (Gillespie et al., 2009; Hansen et al., 2010, Dekkers et al, 2017). The annual cost in production-related losses of PRRS was estimated at $664 million in US in 2013 (+ 18% compare to 2005) (Holtkamp et al.2013).

The PRRSV genome comprises a single, positive-sense, polycistronic RNA strand of approximately 15 kb.

PRRS viruses are classified in two types:

  • The type 1 was identified in Europe (EU) and the type 2 in North America in late 80’s. The two viruses cause similar syndrome, but their sequences revealed significant divergence (Brar 2015).
  • The PRRS type 2 is subdivided in the two subtypes: the North American PPRS virus (NA) and the Hyper virulent PPRS virus (HP-PPRS) also called the Chinese PPRS virus. The HP-PPRS virus is genetically identical to the NA-PPRS virus except for a deletion of 30 amino acids in a non-structural protein (Nsp 2). The HP-PPRS virus was first discover in China.
